I'm curious how River knew that it was the right time to appear and tell the Doctor who she really was. If the Doctor jets off and meets her younger self right afterward, perhaps her older self remembers that he learned about her at this point in his own timeline?

As to the quote - I read some transcript that quoted her as saying: "Of course not" - but not important as I agree with your assumption as to her meaning. Although I am not sure 100% that she is the one who shoots him inside the suit. 99 percent, but not yet sold as to we never see the face shield open till Amy takes a crack at her.
On the point of her knowing when, I would also guess it does have something to do with what she said (can't remember exactly whether it was in SitL or series 5, or just this past year) - but she says when the Doctor meets her she was very young ... not referring to the Demon's runs events, but the upcoming season will prove you right by her remembering when the Doctor come's a callin'!
