Unless you go back to the end of the Bronze Age and Linear B, a Greek letter is just a Greek letter. They are the same today as thousands of years ago (though sound values have changed). Koine is just a stage of the Greek language. Asking if the letters are Koine is like asking if Latin letters are Middle English.
